海外から招聘した講師による英語での集中講義を通じて、特定の専門分野の基礎的な知識および最先端の研究内容について学ぶ。積極的に質問を行い議論に参加することを通して科学の現場での英語でのコミュニケーション能力や国際感覚の育成を図る。

事前に学習する関連分野の論文の内容を理解した上で、英語の講義を理解できるようになる。

授業概要/指導方針 【授業概要/指導方針】
講義に内容に関連する英語論文を事前に読解、学習し、キーワードやキーコンセプトを理解したうえで講義に臨ませる。少人数クラスのゼミ形式の講義と議論に対して主体的で積極的な取り組みを奨励する。

Students will learn about the ubiquitin proteasome system and their overall biological functions in health and disease in human and plants. Special emphasis will be given to current laboratory methods in protein biochemistry, and entry-level structural biology analyses. The short course will integrate active discussion on current studies, as well as practical skills in structural biology illustration tool (PyMOL).

Lecture 1: Intracellular Protein Degradation – Focus on the Ubiquitin Proteasome Pathway

We will discuss the importance of proteins level and cellular homeostasis regulation. The discovery of the ubiquitin system. Modification by ubiquitin, the multiplicity of the ubiquitin code. The ubiquitin system in health and disease (in human, plant, and yeast).
2 12/8 [2] Dr. Nitzan Shabek Lecture 2: Self-regulation of the ubiquitin system Part 1: Focus on the proteasome mega complex and how ubiquitin and other components regulate themselves by the ubiquitin system.
Part 2: *Student discussion: students team up in small groups and each in their turn present and discuss case studies of how their research can be related directly or indirectly to protein degradation and/or modification by the ubiquitin proteasome pathway. The presentation should be 6-7 minutes.
- Students are asked to prepare their topic presentation before the class.
- The teams can be 1-4 students
- The discussion session will take about 30 minutes and the instructor will expand the topics where needed.

* Prior Lecture 2 - students are expected to team up (1-4 members) and pick 1-2 research article that demonstrate a connection between their own research topic and intracellular protein degradation. Each team will share screen and discuss via PowerPoint the research background and main results (6-8 slides and up to 7 minutes).

Part 1:
The era of selective protein destruction in pharmaceutical drug discovery: Introduction to past and current development of targeted protein degradation therapeutics (e.g., small molecules, PROTACs).

o Students are asked to download PyMOL (free trial)
o Introducing PyMOL basic commands (a structural biology graphical presentation software).
o Interactive protein structure interrogation, homology modeling, as well as directed protein evolution (via PyMOL and web-based tools).

The newly discovered phytohormones, strigolactones (SLs), regulate numerous essential growth and developmental processes including branching, leaf senescence, root development, and act as rhizospheric signals for communication with symbiotic fungi and parasitic plants. It has been shown that SL signaling requires three distinct enzymes: D14 α/β hydrolase, F-box MAX2/D3 ubiquitin (Ub) ligase, and SMXLs transcription repressors. SMXLs levels are controlled by MAX2 and the phytohormone receptor D14. Despite these findings, key questions concerning the phytohormone perception and regulation by the conformational dynamics remain unanswered. Here we show that MAX2/D3 has multiple functional states that are determined by distinct structural conformations. We demonstrate biochemically and in planta the key role of MAX2/D3 plasticity in the recruitment of D14, SMXLs, and SL perception. Altogether, this work provides new insights into the regulation phytohormones signaling cascades by the ubiquitin proteasome system in plants.
